Q: Can this work as a replacement speaker for a jbl618s subwoofer?

Yes it is a low pass speaker and it slams hard for the price but it is not a jbl. I beat the hell out of mine with a 4k watt peavy amp

Q: What box would you recommend for two at 50 htz with the trunk open?

I don't about use for in a car, I use for pro audio application in a sound system for a band. You might be better served by getting a car specific speaker system. If you have the speakers already get an empty sub box for a car and try it out, might work fine. They do pump out some great low end, at least in my type of usage.
