maybe 1/2 I would keep it to 1/4

Q: What size cable can be run through this pulley?

1/4 inch should be good you may be able to get a 1/3 inch cable into it.

Q: Is the 331lb the working load limit or the breaking load limit?

Not sure but I am holding a 50lb table with them and put my fat cats on the table and it holds without an issue.
